Isca the Unbeaten faces the following series of challenges, how far does she get?
1) vs. Longshot: Who wins in a game of poker?
2) vs. Cypher: Who translates an obscure alien text the fastest?
3) vs. Quicksilver: Who runs from West to East Coast the fastest?
4) vs. Hulk: Who wins in an arm wrestling contest?
5) vs. Dr. Doom: Who builds a functional time machine the fastest, given access to the resources of Latveria?
6) vs. Worthy Thor: Who lifts Mjolnir above their head the fastest?
7) vs. Classic Juggernaut: They run at each other, who can remain unstoppable?

Isca's ability is just that she can't lose, not that she has to win. She can also stalemate.....
So I can see her stopping at 1, for example if they both get a royal straight flush.

Ok, so lets add a tie-breaker rule to each challenge:
1) If the hands are equal, Longshot wins.
2) If they are equally fast, Cypher wins.
etc.
Basically, if Isca cannot win she loses.

Yeah, my point was that her ability isn't to win, it's just not to lose. But she can still 'lose' in other terms.
Longshot's ability is to engineer outcomes favourable to him. Poker is about betting.
Longshot bets their lives. Whoever has the winning hand, gets to kill themselves. He does this with the probability being superhigh the mutants bring him back to life a few hours later.
Isca is unable to refuse the bet, so she wins. As she herself said, there is also a victory in dying, so....dies at 1.

Not an expert, but isn't there flush tiers? Like hearts best everything else?

Huh, guess not. Thought I heard hearts were the highest suite, followed by diamond, spades, and finally clubs.

Longshot is a hero; he wouldn't impose a death prize even if this particular poker game allowed it. If he'd lose either way he'd just accept defeat rather than spitefully kill the winner in the process.
Killing Isca is a different topic -- it's a very easy thing to do, given her powers. As da Costa and others have demonstrated she cannot refuse or reject any challenge thrown at her way; her powers automatically activate in such a situation whether she wants it or not.
Deadpool could walk up to her and issue a challenge
"Whoever shoots out their brains first is the winner!"
and then try to blow out his brains as fast as he can. Isca would still be faster.

As a side note, isn't Longshot a mojoverse citizen rather than a mutant? Would the resurrection protocols apply to him?

Then he can simply say whatever is in his character - whoever wins this hand, has to retire and live a long and boring life free from all competition , whatever your mind thinks up.
Or whilst you say he won't impose a death prize, then sure - Loser gets to go out on a date with Longshot. Don't forget Longshot's other ability, to attract women - a date with him can be a win for Isca.
Ultimately, my point is the same - she can't lose, which is different to she always wins. There are a myriad ways in which this can manifest, which can work in Longshot's favour.

The mutants resurrected Captain America, most decidedly not a mutant (I thought you'd bring it up). Unlikely that they'll also do the same for Longshot, sure.
Very unlikely. If only someone could make it so he'd just so happen to be on the List to be resurrected beforehand.....
They've also expanded the protocols to include humans.
By the by, anyway, as you say, as the parameters are engineered so SHE dies, not him.

That's not how poker works: you don't get to place a bet that forces your opponent to do something or give up on something, unless he/she agrees to it. As long as he/she can match the bet he/she can continue playing. Since the OP just mentions it's a game of poker for a win, all bets have a value of 0. Isca could then bet whatever she wants in return, such as flashing a charming smile. She does not have to agree to a date or retirement or whatever. And if Longshot bets a date with Longshot, Isca can politely decline and say that she does not need this prize.
Furthermore, Longshot is not allowed to issue additional conditions nor additional challenges; the OP says that the challenge is poker -- nothing else.

As I said, that question was a side note. I was curious if they had resurrected non-mutants, so -- thanks for the info. But it is irrelevant to this topic.

I think her limits are currently unknown but she's likely an omega-class mutant so she's potentially as powerful as full-power Legion or MJJ or the other reality warpers. Of course, unlike the full-fledged reality manipulators, she has very little control over her power which functions passively and automatically.

You are trying to add another challenge. The OP only mentions a game of poker -- not this additional challenge by Longshot of "Whoever wins this hand gets to do X."
As soon as Isca is engaged in the poker game she will win it (since a tie is impossible due to the tie-breaker).
Your "whoever wins this hand, has to retire and live a long and boring life free from all competition" is not a valid bet in a game of poker, unless such a thing has been agreed upon by both players. And, according to OP, there is no such agreement. So if that is supposed to be a bet then Longshot cannot include Isca in it, Isca can bet something else instead. If, on the other hand, that is *not* supposed to be a bet but a new challenge then it's neither allowed by the OP nor possible due to Isca's powers (if it would cause her to lose the initial challenge).

I'd suggest establishing house rules. Players usually don't get to impose bets, the house does that.
And in general make it so the contests rules can't be gamed.

If you mean limits in the sense she's forced to switch to winning teams instead of simply forcing the team to win, then yes.
