Not Symbiote nor Wolverine originated in a game to my knowledge. Either would beat Cloud in a stomp anyway. Combined even more so.

Spider-Man: Web of Shadows has Wolverine being infected by a symbiote to create Symbiote Wolverine.

Does that qualify as an original character?

No because A) comic property B) there is also an actual comic version of Wolverine with the Venom symbiote.
