Where do the Kai and Supreme Kai come from? Are they a special race bred specifically for the job of Kai? Were they once mortal and attained Kai status after being selected for the job?

Was anything ever revealed about their background?

Well as we know all of the Kai's are different species, so I'd go along with gained a special status. Definitely in the same way these Gods are attaining their namesakes
