Spider-Man gets one punch on Flash.. A free shot and Barry gets a free shot at Peter..

Flash goes first and gets one free punch at Peters face. Does Peter get up or is he knocked out cold?

Peter then gets free punch at Barry's Chin.. Does Flash get up or is Barry out cold?

I assume they can't use their powers?

So for example, Barry can't vibrate to take the punch? Because otherwise, he's just a human..

Depends on who goes first. Both character's if they were to take a sing at their max would decapitators their opponents head.

Ding ding ding. We have a winner. It all depends on who goes first, especially if characters that can vibrate are not allowed to. Thus it is all offense and no defense.

If Spider-Man goes first, he literally takes off Barry's head and it falls to the ground a couple hundred feet from his body. If Flash goes first, he literally takes off Peter's head and it goes into lower-earth orbit for the next couple decades or so.

If any part of Flash was moving at lightspeed, even a fingernail, and it connected with Spider-man's head, it wouldn't just knock his head off, There'd be so much kinetic energy involved, Spider-man would be instantaneously vapourised. It'd make for a crap comic, but Flash would win the as per the OP stips, no question.

Doesn't matter. The speed force is not something Barry turns on and off. The SF is always there to protect the Flash family.
