Vamp is more than enough to kill Wesker.
Not only his regen powers are more impresive, unless you give him the nanosupressor shot he invulnerable also he was overpowering and taking direct blows from Raiden, and we all know the strenght levels of that dude.

Vamp's regeneration is no where near Wesker's either. We've seen Vamp got down after getting shot once on at least 3 occassions. Meanwhile it took dumping Wesker into a volcano and using 2 RPG's to finally kill him.
We saw Vamp fight Raiden but Raiden always used swords and knives against him, never full blown punches. Wesker could easily heal from stabs and lacerations.
Never mind the fact that Wesker is an actual bullet timer and Vamp isn't. Vamp doesn't have a single advantage going in this fight other than his ability to read muscle movements. Which won't help against someone who's massively stronger and faster than he is.
Big Boss won't even be a factor here, Wesker will probably kill him off the bat.
Well im going just with was present in the games, Vamp was holding and overpowering Raiden and he stabbed himself to break the hold and Raiden actually was giving him blows in the Rex fight,i see him doing well if people like Chris and Sheva can handle Wesker just well, dodge his attacks etc.
And Big Boss would play a role, again, if a total inferior character like Chris can handle him well, i see Boss giving him fight, in PO he fought a even faster character with mental altering powers in Gene

Yeah, the writing is a very inconsistent in that game and the series in general. For example Raiden was able to hold back Outer Haven, but earlier in the game before he fought Vamp he was being held by 4 Gekkos and Solid Snake had to save him by shooting out one of the cables.
As far as Raiden actually hitting Vamp goes, it just shows that Raiden's actual striking power is pretty abysmal. I remember in their first fight when Raiden threw his sword at Vamp, Vamp dodged it and the sword ended up hitting a Gekko, and it barely went through it's armor.
There's no evidence of Gene being faster. Even if he was comparable, Gene has already shown that using his speed drains him and leaves him vulnerable. That's the only reason why Big Boss beat him, which won't apply to Wesker since he doesn't tire out as fast as Gene does, and is massively stronger than Gene is.
Also Chris is probably physically stronger than Big Boss and at least as fast. Not that Big Boss himself has any impressive speed feats.
Also Chris didn't handle Wesker. Everytime they fought Wesker kicked his ass until the end of the game, and Wesker still survived being dumped into a volcano and took 2 RPG's to finish off. You make it sound as if Chris beat him on his own in hand to hand.
Big Boss smokes a cigar from a distance while Vamp owns Wesker.
Wesker is nothing compared to the shit Big Boss and Solid Snake take on a regular basis. They are like the Captain America of video games. Somehow beating the shit out of anything that stands in their way.

Big Boss and Snake get their asses kicked on a regular basis. Big Boss has been taken down by Volgin, Cunningham, 4 soldiers etc. and he's going to get his ass kicked by Skull Face in Phantom Pain when it comes out next year.
Snake has been taken down by Vamp, Ocelot, and even Solidus was too fast for Snake to keep up with.
You can throw Snake in this fight and Wesker would still tear them a new A hole
