tl;dr
So long as at least one 'slice' is alive throughout the multiverse, Maker himself cannot be permanently beaten -- each slice will just keep pressing forward with the same collective mission/goal that Ultimate Reed's had since the beginning: shaping the "perfect world" (albeit in their own unique ways.)
This was also shown when one avatar was killed after Leech(who can nullify the superpowers of ANYONE in his vicinity now?) gimped his abilities, and then a different avatar just immediately spawned-in and took his spot:
