My vote is simple...and it is for the most badass character I have ever experienced. The one character who grew old with me and felt human besides his superhuman qualities.

Yes, he's simple. Yes he's not very exciting, but you know what, he's not the most copied video game character ever for no reason. He doesn't need a deep story, a deep character bio, or any of that stuff. Ryu has stood the test of time because of the stuff he can do in the game. Overall, not the deepest character ever, but he is epitome of his genre and represents like no other character ever has. He even inspires 'real' martial artist.
This guy has thrown hands with Kyo Kusanagi, up to Galactus. Ryu's the best because I love fighters and he represents what it really means to be a true warrior.

Gabriel Knight - Voiced by Tim Curry he is a New Orleans secondhand bookshop owner who is investigating a series of Voodoo related murders.
He is the last in line of Witchhunters. Schattenjagers who kill werewolves, vampires ect.
With 3 games under his belt this womanizer and funny guy is certainly up there.
Agent 47 - This guy kicks ass. A genetically altered assassin with no soul who can enter the most secure locales and eliminate his targets with nobody knowing he was ever there.
Has Mario ever actually been given character development? Like, if it was "random Portuguese carpenter" or "Mustachioed Navy SEAL" instead of "Italian plumber" would the games be much different?
Same with Sonic, for that matter. The TV series gave him some personality, but the games certainly haven't.
If anyone's familiar with the baseball stat "value over replacement," I don't think Mario's or Sonic's is very high.
Mario isn't actually italian, or a plumber for that matter, since those aren't really canon, as far as the word "canon" actually means anything in Mario. The real canon being that Mario and several others (including Donkey Kong and Bowser) were "Star Children," and are said to posses some kind of power. As established by Yoshi's Island and Partners in Time, Mario's lived in the Mushroom Kingdom all his life. He just dresses like a plumber for some reason, though in the original Donkey Kong he was supposedly a carpenter or construction worker, before everything got all confusing.
As for personality, no, he doesn't really have one beyond your standard heroic bravery and kindness, and such.
Oh, the games give Sonic a personality. Especially the 3D ones. Few examples:
