Thanos w/power gem can take out some pretty powerful beings, but would it be enough to take on the all mighty TYRANT? Or can Tyrant even still, put the Titan on his wallet?

Thanos, probably. He's one of the few who can tap in the full power of the gem.

Thanos was nearly killed by Thor when he only used the power gem.

Also you do realize Thanos was taking on more than just Thor.
