If Joker got his hands on the reality gem, could it give him the power to beat Jenny Quantum?

Yes.

This isn't Joker's first rodeo with abstract-esque/godly power. He knows how to wield it extremely well. In fact, has been stated in a few different arcs (Emperor Joker, Rock of Ages, and Trinity) that Joker's mind adapts to infinite levels of power far easier than most others.

The issue though, is the Reality Gem truly infinite in power?

Jenny Quantum is theoretically universal in her reality warping powers, as she was able to create that flying turtle island dimension thing for the Doctors and past Jennys to converge and live in.

So would just one gem alone give Joker enough power to beat her? Without something like the Power Gem backing it up?

Next to that level of power, the reality gem certainly wouldn't present an overwhelming experience(especially by itself.) As I always understood it, was that the power gem was the OOMPH behind the others to make them universal in scope.

How many other trans level characters are capable of designing pocket dimensions from scratch?

All the Gems (alone) are potentially "universal." (or even better?)

Warlock erased a Timeline with the SG.

Rune froze the Time-stream with the TG. (only the LT was un-affected)

Eternity/Infinity stated that with time,
PG/Thor would become a threat to a good portion of "All That Is." (multiverse?) holy!

... those are a few examples.
