Like in Dante's case that's a question of distance. They both have rather good SPRINTING speed feats (not sure about Samus' distance feats) and CHEATS like Quicksilver in Dante's case (or Samus' phasing). But this is a marathon race being banned, and so is flight.
That's how I understood the stipulation and why I think that's basically between Speedy and Sonic with others deeply outclassed by those two.

As for distance in Super Metroid Samus runs a very long way, smashing through cliffsides and a spacecraft hull when she does so, IIRC.
Rainbow Dash has her outclassed in speed, but she needs to fly to do so. Being higher up essentially puts her in a lower orbit and makes her travelling path longer, and Samus can shoulder barge through the planet's hills and stuff.
I honestly underestimated Sonic. Didn't realize base Sonic was FTL, that's ... Unfortunate. And It's been a long time, so I'm a big forgetful of Speedy.

If flight is on, then I see Dante (space flight in fight with Mundus) as well as Rainbow Dash attempting to cheat by "suborbital jump", basically shortcutting this way.
Though anyways Sonic by this time stands on finish line and lol's at opposition.
And IF Quicksilver is allowed...but Sonic has beaten time manip so this way of cheating is meaningless and all hail hedgehogs!
Samus has the speed booster, which allows her to hit hypersonic speeds and break through obstacles. She can also shinespark, which is essentially just launching herself in a direction and destroying everything in the way.
That's right. A good sprinter-burst-speed, same as Dante. I can see them both in a race with Rainbow Dash for a bronze which is going to be damn interesting thing...and may quickly degenerate into 2 combat behemoths on one poor lil'pony.
Because of unclear "measurements" (unlike Dante's case where clear calculations were made)? I think we have a winner and a loser, thus (based on those videos Samus is MUCH slower than Dante even without Quicksilver and Sparda DT feats).
So it's
1) Sonic
2)Speedy
3) Rainbow Dash
4) Dante
5) Samus
Unsure about 3 and 4 (but everybody love ponies) and flying OVER obstacle is IMO much greater shortcut than running through it.
Samus has few other feats that put her in the hypersonic range, such as crossing a few hundred feat without being seen and outrunning gunfire while doing so.
Based on "running around Mexico in second" feat? Other than this, Samus is clear last (space jump IS a shortcut if handled properly, with reentering the atmosphere thousands of miles below, however this is applying RL spacecraft logic to her reentries). And the race for silver and bronze is epic (everyone agrees that gold is Sonic's).
