No, in his armor form Colossus is anything but human. Essentially he just a big pile of metal just like Clayface is a big pile of clay. I don't think the possibility that Colossus might have a metal heart that is somehow pumping metal blood through metal veins really makes a difference.
In comics, being alive = having a lifeforce imo.
There are numerous characters that lack internal organs and yet are just as alive as anyone else (classic Chamber, Silver Surfer, etc)
