A very long time. I remember hearing that he first thought of it when he visited the ancient Martians (before they were wiped out) and found out that they believed in a "life equation", so he figured there must be an opposite to it.

Still,Darkseid would have to have been looking for the equation for over 50 000 years,considering Jonn was alive on mars atleast 40 000 years ago.
