Spider-Man's physiology is enhanced such that he has the proportional strength of a spider either due to a radioactive or genetically engineered spider's bite. I don't know if they've ever elaborated on how his physiology is altered exactly.
Real world logic applies more easily to Wolverine because he is still a human, but with the genetic gift of accelerated regenerative processes.

I have to admit I haven't read the comic you speak of or I have and I've completely forgotten it. This was a blood cell? Was this magical energy or physical energy?
But perhaps that amount of energy is enough to regenerate any organism from a single cell not just Wolverine. Every viable cell has the entire genome, except red blood cells in which case regeneration would be pretty impossible.
