My theory:

The radioactive liquid he injected himself with when he first defeated Morlun is killing him. Peter thought it had been purged from his system, but he was wrong. Eventually, when it will be purged, he will be purged of ALL radiation. Including the radiation that was passed on to him from the spider that bit him and made him Spider-man.
Now, remember that Ezekiel told Peter that the spider that bit him was a mystical spider, which was trying to pass it's powers onto someone before the radiation killed it. So, if the radiation was bad for the mystic spider, then I'm betting that it's bad for Spider-man as well.
The radiation in his blood is a liability and a weakness. The same way as the adamantium in Wolverine retards his healing factor from acting on it's full potential. The radiation that's been within Peter all these years that he always assumed was just a natural byproduct of his powers, is keeping him from being what he would've been if the mystic spider hadn't been dying.
When the radiation is finally purged, Spider-man will evolve into what the spider that bit him truely wanted him to be.

Why? He's not the Hulk. The mystic spider bit him to give him spider-powers. It's bit would've given him his abilities w/ or w/o the radiation. So, what purpose does the radiation serve? Clinging to wall? Nope. Spider-sense? Nope. Webbing? Nope. Agility? Nope. Strength? Nope. There's no reason to believe that he would lose even one of his powers even w/o the radiation in his blood. He has "pure totemistic" powers.
actauly the radiation could of strength some of his powers kinda like wolverine thou it weakeness his helaing factor it allows him to do certain thigns he would not normaly be able to do.
my theory which is based of urs is that the radiation made him faster then he normally would of been so when they take it away i think u will find he not gunna be as fast.

That is a top theory dude!! It makes sense tbh because from the solicitations it seems that Spidey will be going on a supernatural journey of sorts on the brick of death... so I'm guessing that the theory above will show up in some way, shape, form in the issue.
Looking forward now to the conclusion of THE OTHER
although still tentative about how his eye will be replaced, I'm sticking with a temporary boost of healing in the cacoon... if he even goes into one.
