Well, I think there is now a direct connection between the Plagas and the T-Virus. In an interview (posted by me on the previous page of this thread) that they will go to the source of the T-Virus, which came into existence at the place where human life began, and as seen by the trailer, they did look a lot like Plagas.
So origin place of T-Virus, Plaga like people.
Of course, the actual T-Virus was created in a lab by stabilizing the Progenitor Virus with leech DNA.
Also, if I remember correctly, it's recent volcanic activity that awakens some "long dormant virus" in this game. Which would seem to indicate that the virus was underground originally, and as RE4's epilogue showed, the first Plagas remains were found underground in the mines.
So maybe this is some older virus, that later evolved into the Progenitor Virus and the Plagas? That fits learning the origins of the virus, and shows a connection to the Plagas.
GameTrailers analyzed the trailer I posted earlier and noticed quite a lot of interesting things: http://www.gametrailers.com/player/34711.html
Some RE5 footage I found to be quite cool:
Honestly, alot of peeps may not play this game all the way through. For the 1st time ever... a game looks too scary. It's gonna be like, "Dude I can't finish this game, too scary" Or...
"Hey man, let's play some RE5".
"What time is it?"
"About 10."
"We can't!"
"What, why?!"
"We can only play it in the day time!"
"Pussy..."
Seriously tho, I know peeps who were like that wit RE2, so I know there gonna be scared shitless by this game, hell lookin at the previews I think this game might make me a lil alert.

I think the game looks fantastic, but at the same time it looks too much like RE4. I know thats a good thing, but it seems like every single animation is just reused from that game.
In this vid they actualy bring up that point of it being similar to RE4, they go as far as to call it more or less RE4 in HD: http://www.gamevideos.com/video/id/20174
