Not with the director Sam Raimi, with the stars Tobey Maguire, Kirsten Dunst, and James Franco.
There may be a few more movies in a few more years, but I think this will be it for a while.
The first came out in '02 which means they've been working on them for probably close to 7 years by the time Spiderman 3 comes out. (If you figure that most effects heavy movies take 1.5 years to make, and that the film shooting of the actors comes before that.)
The director and these actors are not Roger Moore; they aren't going to continue spending the time and energy that could be used on growing their careers to cater to the fans of a franchise.
I find it hard to believe that a studio would finance a picture that would cost as much as the next Spiderman movie (and it would cost alot, since they would have to either raise the bar on sets, action, and effects, or at least produce the same as what was in Spiderman 3) cost them, when the director and main characters that people went to see will not be in them.
Especially when the story has been completed, the character arcs finished, and a great trilogy has been made.
Sure, there are many more stories, but nobody will want to go watch some new guys facing the threat of Rhino, Electro, or Mysterio.
Nobody except for the comics geeks, but sadly, those guys only make up about 10-15% of the movie's audience. Most people that see the films know little to nothing of the comics, and they will be done with Spiderman stuff after this film ties things up.
We may see some more films as a continuation or a reboot later on, but I'm betting on 5-10 years before it happens, perhaps longer.
