Batman gets the coolest lines.
For example?
Random Villain: "B..but I saw you crushed under all that debris, how did you survive?"
Batman: "I'm the Batman."
I also like a lot of comments from the ultimate Universe eg Cap's "You think this stands for France?" moment. I loved it when Ultimate Xavier took down Magneto with "Goodbye old friend, give my regards to the Dodo."
My favourite line in a comic probably has to be In the last issue of Ultimates vol 1 when Iron-man gets the idea of disposing the universe destroying bomb by giving it to Thor to get rid of and Black Widow says something like.....
Black Widow: "What's Thor going to do? Dump it in Narnia, we aren't talking about a Norse God, Thor's a delusional schizophrenic."
Then the doors open to reveal Thor stood silhouetted in the sunlight....
Thor: "Oh ye of little Faith."

Tommy Monaghan and John Constantine always get great lines.
Swamp Thing is fun to listen to, especially if you're reading the Alan Moore era: I like reading his lines aloud: "The Green...all your....hate...and...anger....washes...over into it....the Trees...The Trees!...THE TREES!" if you read his lines aloud with the ellipses in them, at the pace you imagine Swamp Thing talking in, it's great.
Jack Hawksmoor always gets good lines.
Batman is key for some great badass lines- "I'm not here for the wedding. Just the fight."
Dr. Strange always gets good ones too. He works best when he's more witty and literary than when writers make him sound topical though.
Hands down though, Blaastar from the Negative Zone has the best lines.
