Most of the arguments aroung Destiny and such are distractions. With control over Time, Reality, and the Soul, Thanos could transform the very nature of existance to the point where the concepts that the Endless embody have no meaning or are no-more/never-were, or whatever.
However, this would be more-or-less a suicide attack, as Thanos would have to sacrifice his own Endless qualities as well. "Victory" would come at the cost of his own Deams, Desires, and so on.
