Mxy has already defeated a fully powered Spectre. The only difference is if the Spectre is backed up by the Presence. If he is, he is only second to the Presence.

The Spectre would blink him away, don't remember him defeating a Fully powered Spectre ever. While he was hostless the Spectre was tearing through magical beings left and right and a side effect to those battles was Mxy losing most of his magic, even a hostless Spectre should be able to win..
