Surprisingly easy, when you stop and think about it.
The movie has to have at least TWO actors who have played comic book characters in other movies. Like...

Batman (Keaton) and Superman (Reeve) first teamed in the live action movie, SPEECHLESS.

Sebastian Shaw (Kevin Bacon) and the Joker (Nicholson) were in A FEW GOOD MEN.

PULP FICTION saw Nick Fury (Jackson) and the Abomination (Tim Roth).

The Penguin (DeVito) and Mr. Freeze (Ahnold) in Twins.
The Penguin (Metedith) and Judge Dredd (Stallone) in Rocky.
Agent Coulsen and the Riddler (Carrey) in...Mr. Popper's Penguins.

The STAR TREK reboot had the Hulk (Bana), Gamora (Saldana), and Dredd (Urban).

IMMORTALS had Whiplash (Mickey Rourke) and Superman (Cavill).

And so on...!
