IMO, real life Fisk is bigger, stronger, and more skilled than the animated Joker, but animated Joker has the advantage in pain tolerance and has a butcher knife...

In my minds eye, this is a balanced fight; atleast balanced enough so its not spite against either character...

Using a live action character vs a cartoon almost always means spite vs the live action one. Because as strong as Fisk is, he's probably average or slightly less in an animated world.

And even Fisk's suit won't do him much help, as Joker will probably try and smack him in the head with the blade.
