The gem did amp him, using a gem offensively makes it shine, he used it to hurt Thor when Hulk attacked. So he tapped alredy into it, hence the reason he was stronger than Hulk. In the next movie I expect a rematch, Thanos without the gem vs Hulk.

He was actively usingit when Hulk attacked him. Also, in some instances Thanos used the gauntlets power and the stones didn't glow to my recollection. The feat is ambiguous at best if you want to favour thanos. He was either still amped because he used it against thor or it amps him passively, like in the comics. Maybe we will see Hulk vs Tanos w/o the Gauntlet in the next movie.
