Round 1: No. Even if he somehow doesn't get pierced by the vibranium-tipped horn, which is a major if (though, to be fair, nothing has managed to actually pierce Luke since the power-up), the concentrated blunt force impact as well as being sent flying dozens of meters and crashing into a boulder is going to rock him hard. Because while he has taken hits like the following without damage before, the force was much more spread out than the rhino's horn would be:

And I don't actually know what the penetration power of a missile fired from an FIM-43 is. Though Iron Fist can punch through an armored truck, so there's that. But either way, the blunt force alone will hurt him.
Round 2: Yeah, Luke could take the win. He just needs to avoid the horn and could literally punch holes in the numerous exposed parts of the rhino's body if he really wanted to.

I kind of wonder how Cage's movement speed compares to the rhino. Because he can cover a 40m dash in 3.72 seconds from a stationary start. It's easy to work out the average speed over distance, but that doesn't actually give us the peak velocity he'd need to hit to average out to that number (considering he's starting from 0). And then there is the fact that many sprinters only hit their max velocity around the 60m-80m mark in a 100m dash. And I suck too much at math to work it out quickly lol. Plus I am too lazy to actually put any real effort into it.

This sounds about as good as it gets to me. Off the top of my head I know Rhinos can easily run faster than a human and considering their weight, that is beyond impressive . They are known for flipping cars in Africa with casual ease. But he should be able to punch holes like you said though. He wants no part of that horn. It would severely injure him I'm sure
