Sorry, but you're exaggerating. Where exactly are those major changes ? For the record, I consider the whole organic-web thing as a minor, not that important change.
Something else : there are people who are not afraid of mentioning Stan Lee himself, as to prove their point and to claim that they - and nobody else - are the true Spider-Man fans and that storywise absolutely nothing can be changed. According to them, Stan Lee had this "grand vision" of Spider-Man when creating him, and those greedy retarded hollywood suits raped his wonderful ideas.
Nonsense. If you're a real Spider-Man fan, you at least should know that Spider-Man was not meant to be made in series. The story was just a single event, nothing more. But it was so successful, that the publisher asked Stan Lee for more stories about Peter Parker.
And Stan Lee did NOT expect that. He knew the story wasn't that bad, but he never could have known it would have such a tremendous impact on the comics. A couple of months later, when the sales figures came in, he had almost forgotten about Spider-Man !! (his words, not mine).
What I'm trying to say, is that some people think the creation of Spider-Man was part of a gigantic brilliant masterplan, and therefore nothing, not even the tiny details, can be changed because "it's all part of the plan and now that they've included organic webshooters, everything is ruined."
No. The first Spider-Man comic was a lucky shot, and Stan Lee did not spend weeks and weeks to deliver the perfect story. It was just a superhero story that was not too bad, but he did not have a vision.
To end, a short "review" of the Spider-Man movie by Stan the Man himself :
"I LOVED IT," Stan replied without a moments hesitation. He then continued, "I loved it as the creator, but I also just loved it as a fan. Purely as a fan watching the movie, it was so exciting. I only saw a rough cut, but it was phenomenal."
Stan also had great praise for actor Tobey Maguire, who Stan has been quoted as saying might not have been his first choice - until he saw the young actor's performance in the film. "[Tobey] was just so natural and so believable. He seemed like he wouldn't be the first guy the girl's would chase around; maybe a science major."
It's funny when you think of it : Stan Lee is much more broad-minded and easygoing about the Spider-Man movie than most of his fans...
