New map, that is larger than all of the cities in AC2 together. The teams working on AC went into insane amounts of detail in making sure that the cities in-game matched their historical counterparts; this means both sending teams out to get thousands and thousands of photographs, and studying historical maps of the time period. Then they had to model all of the buildings and texture them. As the buildings are very different than the ones in the cities in AC2, they are clearly new assets. This takes a very long time. There is also a much wider variety in what is seen in Rome - they had to create all of the ruins as well.
Almost every character is remodeled. Ezio has new robes. Civilians look very different than the ones in AC2. Guards mostly look the same, but the Papal Guards are new.
New weapons requires new scripting, new models, and new textures.
Assassin recruits require new models, new scripting, and new AI.
Voice work takes a long time.
Kill streaks and executions require new scripting and programming, not to mention new animations.
The Lairs had to be built, the puzzles created so that they offer a level of challenge but balanced as to not be too difficult, new models created and textured, and scenes scripted.
New memories, which require scripting, AI, voice work, animations, and so on. Programming to unlock Cristina missions as your sync rate increases. Programming to change the AI of the thieves, courtesans, and mercenaries as your rank in the guilds increases.
The multiplayer, which...yeah.
That is an insane amount of work that had to be done. And I know I'm missing a lot.
