Okay... I didn't think this would need that much clarification but...
All the participants lose their superhuman abilities, from the Canary Cry to Wonder Woman and Thor's prodigious strength. They also lose any externally derived attribute enhancements, be they adamantium skeleton durability (or poisoning 😐) or Midnighter's fight implants, heart etc. They also lose weaponry (including the physiological e.g. claws) and the advantages their costumes would afford, vibranium means nothing, kevlar means nothing, Aegis forged bracelets mean nothing.
They are all still essentially perfect human specimens, with whatever inherent advantages their respective builds would entail, be they the strength of being a six foot something Norseman or the agility of a comparatively petite half-Asian girl or the slightly heightened sense of smell of a blind man.
But while Batgirl would no longer be 3× baseline strength and 5× baseline speed she would still be a strong, fast and probably most importantly skilled woman. While Wolverine and BP would no longer be superhuman, they would still be strong and fast. While Deathstroke etc.
(They'd still probably be stronger, faster, more agile etc. than real world human analogues though.)
They would all be instantly aware of what they've lost, but are given no time to adjust to it, so Wolverine, Deadpool or Thor may charge into the battle, but they'd have to be idiots to do so knowing that they're no longer incredibly durable or can't just heal.
The arena is something along the lines of the Colosseum, they are all going for the kill but they are still rational, ergo they can team up if you think that they would.
