I think Harry. He's got more passion for fighting and is more level headed in a scrap. For example, I don't see Hermione doing nearly as well in a duel against No-Nose.

Granger is more intelligent and schooled in magic arts

Beyond what King said, Harry is also far more talented at combat magic (Defense against the dark arts).

That doesn't really matter. Voldemort was a genius and Harry beat him. Voldemort was the most powerful dark wizard in history and Harry beat him.

He's faster on the draw than Hermione, more willing to hurt/kill his opponent and just a better fighter.

Seeing as how "Expelliarmus" is Potter's go-to... Harry takes this.

Also, the building of Hogwarts reacts to Harry's personality in a good way, he's got help there too.

In this hypothetical scenario, neither are holding back. That means Granger won't restrain herself.

Rowling actually explicitly stated that, books 1-3, Hermione was the better duelist, but Harry surpassed her by book 4 and only got more and more competent.

He's arguably even better in the movies, and he managed to seriously challenge Voldemort. He wins.

To answer your question though, yes, she could. With prep I could kill Harry Potter. I'd use that prep to procure a gun.
