I love the designs, and some of the casting(like Biel as Black Cat, and Russel Crowe as Kraven) is absolutely phenominal, but I disagree with some of the casting as well(Marsters as Vulture, Stormare as Electro, and Campbell as Conners(Baker's already Connors)).
For Vulture, I'm not sure who I would cast, and for Electro, I'd go for Dennis Leary.
But now, I want Kraven in the movies after seeing that Russel Crow picture. It's like he was made for the role.

But you have so many other good villains besides Mystero like Lizard, Vulture, Electro, Hobgoblin, Carnage, Venom (Returning) as well as many more.
I really don't want Carnage in a film. Like, really really really don't. I think he's such an awful character.
From this point, they could do a number of villains successfully.

I think Mysterio could work either as a secondary, co, or starter villain. Main, and only, film villain? Not sure how that could work. Maybe if they took the Kevin Smith/Daredevil approach, but that's Daredevil.
You know who would also make a good Kraven besides Crowe? Gerard Butler. They'd both be fantastic, and both have that gruff hunter look Kraven is famous for.
I had some ideas.
First one:
The Lizard is created due to Connors' constant use of bio chemicals on his missing arm, and begins killing any mammal it sees. As Spider-Man tracks him down, unaware of its connection with Curt, the world famous Kraven the Hunter comes to hunt the Lizard. However, his plans are a bit larger than anyone realizes. Empowered by a mystic formula given to him by an old witchdoctor, Kraven plans to set in motion a means to capture the Lizard and Spider-Man, who he believes to be the ultimate prey.
Setting off numerous accidents to attract them such as releasing zoo animals and creating runaway trucks, Kraven ends off pissing off the Lizard, who now wishes to use Connors regeneration formula to destroy molecules instead of adding them.
Spider-Man soon figures out who the Lizard is, and makes a formula to reverse the process once and for all. So soon the fight of the century begins: Spider-Man vs. Kraven the Hunter vs. The Lizard. With New York City, and possibly the world, in the balance, Peter must defeat these two opponents, save Connors, and stop a crisis that threatens all that he holds dear.
The film ends with Kraven, barely alive after being mauled by the Lizard, hauled off to jail swearing vengeance on Spider-Man for his loss, and Curt turning back to normal, but leaving New York City and his family to find the man within himself, and to make sure that the Lizard will never return to hurt anyone ever again.
Second idea:
The film begins with the police chasing two criminals across the city. One of them, covered in shadow, hides in an abandoned Oscorp building. But, while in there, he discovers a preserved hideout of the original Green Goblin. Learning how to use the weaponry, using the formula on himself, and redesigning the costume, he becomes the menacing Hobgoblin. With this identity, he seeks to gain the power that he has always wanted.
The other crook is Max Dillon, an electrician who barely has enough money for rent. He breaks into the city's powerplant in hopes of hiding out there. However, he falls into the generators, and is presumed dead.
At the Daily Bugle, Roderick Kingsley becomes a large funder for the Daily Bugle. For some reason, he supports Jameson's hatred for Spider-Man, and wishes to contribute to it. After an argument with the two wealthy spider haters, Peter is fired after expressing his opinions on the subject. This leads to Peter getting a job at his old high school as a teacher.
During this, a body arises from the electric generators. Max Dillon, now granted incredible powers involving electric manipulation. He stays low in hopes of being away from everyone, but is found by his old friend who is now the Hobgoblin. Now revealed to the audience as Roderick Kingsley, he persuades the mutated villain into being able to be remembered. Making everyone proud after being such a disappointment to those around him. Electro sees this opportunity, and agrees on a team up to kill Spider-Man. Roderick makes him a costume as well, though it is mocked by Spider-Man for the rest of the movie.
After an Electro battle with Spider-Man(with Pete being sent to the hospital), Peter must face the two villains together, and will do everything he can to win against these super powered psychopaths and save New York City.
Can Spider-Man win? Will the Goblin Dynasty be ended once and for all? Will Peter be able to retain his new job?
And in the whole mix you can add the Peter and Mary Jane love story anywhere you wish, with them getting married at a certain point.

How about just Mysterio and perhaps Lizard for 4, since Mysterio is the only impressive villain left bar from Chameleon.
Not to beat the fact that Mysterio is awesome and one of the oldest and best villains of the Spider-Man franchise, he is the No. 4th greatest Spidey villain.

There are a LOT of villains that would work perfectly in a Spider-Man flick, many that could be taken more seriously in a film than Mysterio.
