As much as I love The Simpsons, I don't think it's best anymore. The old seasons are still great yes. The newer ones recently have been decent too. Personally I like South Park best, but that's just me. I also like The Boondocks too though. But if we're going old school, it's always going to be Dexter's Laboratory for me.

Best American cartoon OF ALL TIME?
Now that's a harder decision.
There are a few other American cartoons I consider gems in television history.
Batman TAS, Gargoyles, Avatar: The Last Airbender, Ren & Stimpy, etc.
