From the marketing. Batman, Joker are the most popular characters in DC. Harley is up there too. Way more popular than the transformers name before the movies came out. Even after the transformer movies came out Batman and the Joker are bigger names and SS was marketed showcasing them.

Most people I know don't even know who Harley was. lol. Marketing, they scaled back Batman/Joker for the most part. The marketing HIGHLY focused on the team, not Batman/Joker.

People I know who asked me about this film to a man assumed that Joker was the main villain since he's been featured in so much of the ads as well as the music videos and media coverage about Leto's pranks on set while getting in character for Joker. Joker and Batman are known commodities to the viewing public at large, particularly in a post The Dark Knight/Heath Ledger world. It makes good business sense to showcase Joker like WB did, but it's some used car salesman level ethics considering what you actually get in this film.
