From the films opening scene he already has the Power Stone, and by the time the opening title plays he obtains the Time Stone as well. But wielding two Infinity Stones does not make him the most powerful villain in the Marvel Cinematic Universe. He already had that distinction before his hunt for the stones because of his strong will.
Better than Hela. More powerful. Most powerful MCU villain to date.

A single lightning blast did not leave lasting injuries. That does not mean Thanos can take multiple lightning blasts without damage, since he was unable to tank a single lightning blast. Just in the same way that a single punch from Thanos did no permanent damage to Hulk but Hulk was unable to take multiple hits from Thanos.
